ADAPTUS contains the active ingredient tadalafil. It is used to treat erectile dysfunction, which is the inability to achieve or maintain an erection sufficient for satisfactory sexual performance. Erectile dysfunction is a condition in which a man has difficulty achieving or maintaining an erection that is satisfactory for sexual activity. Tadalafil belongs to a group of medicines called phosphodiesterase type 5 (PDE5) inhibitors. It works by relaxing the muscles in the penis, thereby increasing blood flow into the penis when sexually aroused. This effect is beneficial for men with certain medical conditions. ADAPTUS is usually taken once or twice a day, depending on your doctor's advice. It should be taken on an as-needed basis, at least 30 minutes before sexual activity. It works by helping to improve blood flow to the penis, which can help to achieve and maintain an erection.

Like all medicines, ADAPTUS can have side effects.
ADAPTUS belongs to a group of medicines called phosphodiesterase type 5 (PDE5) inhibitors. PDE5 is responsible for breaking down cyclic guanosine monophosphate (cGMP). cGMP causes relaxation of the smooth muscle in the penis, which allows blood to flow into the penis. This helps achieve and maintain an erection when sexually stimulated.
Whether you can take Tadalafil at the same time as your other medications will depend on which medications you are taking.
You are required to complete a short medical consultation questionnaire that asks for any medications that you are prescribed from your GP and will indicate the medications that can’t be taken alongside Tadalafil.
There are several medicines which should not be taken concurrently with Tadalafil. Medicines which can have potentially serious interactions with Tadalafil include :-
• Any alpha-blocker medicines e.g. doxazosin (Cardura), alfuzosin (Xatral), tamsulosin, terazosin, or Flomax, that you can buy in the pharmacy. (These medicines may be used to treat prostate problems or high blood pressure).
• Isosorbide mononitrate, isosorbide dinitrate, or use a GTN spray or tablets
for chest pain or use Rectogesic ointment?
• Nicorandil (Ikorel), used to treat angina
• Cimetidine, used to treat stomach or duodenal (peptic) ulcers
• Theophylline, used to treat asthma
• Antibiotics:- Clarithromycin, Erythromycin and Telithromycin
• Anti-fungals such as itraconazole and ketoconazole
• Seizure medication:- carbamazepine, oxcarbazepine, phenobarbital, phenytoin and primidone
• Heart or blood pressure medication:- disopyramide, amiodarone, quinidine and procainamide
• Anti-viral medication:- telaprevir, atazanavir, retonavir, indinavir, saquinavir and cobicistat
• Recreational drugs:- amyl nitrate (poppers)

How to take Tadalafil : You need to consult your GP before taking Tadalafil. You will be asked to state what treatment can’t be taken alongside Tadalafil and how much one treatment is safe for you to take. You should include: a full stomach, very high blood pressure or a heart condition. A family history of eye conditions such as retinitis or retinitis oculoasal (suddenly severe, eye pain can occur). You, or your child, is unlikely to be eligible for treatment if you are at high risk of serious eye problems because the eye ointment can be applied directly to the eye (photosensitivity). Tadalafil can cause a serious side effect if you are taking other medicines that:- Can also be used to treat certain cancers (e.g. users of doxazosin (Cardura]) can be switched to Tadalafil after use … … … or when needed (e.g. after a coronary procedure or after a stroke)You should not use Tadalafil if you have already taken this medicine and you have a history of severe cardiovascular problems or if you have a history of heart problems such as heart failure.

The aim of this study was to investigate the association of tadalafil and pde12-phosphohydrolase 1(PH1) polymorphisms with the risk of heart failure in Chinese patients with chronic renal failure. We evaluated the associations between the genotype of the polymorphism of PDE-1 (TGACTTCCTGGGTCAATG) and the polymorphism of PDE-1 (TCAGCTTCCGTCACTCG) with the risk of the CHD risk in an Asian population. A total of 119 cases and 101 controls were enrolled in this study. The frequencies of the polymorphisms of PDE-1 (TGACTTCCGTCAATG) and PDE-1 (TCAGCTTCCGTCAATG) were compared with the general population based information. The odds ratio (OR) and 95% confidence intervals (CIs) for CHD and CHD-related death in patients with TGACTTCCGTCAATG and PDE-1 polymorphisms were significantly increased by the TGACTTCCTGGGTCAATG genotype (P< 0.05) and the PDE-1 genotype (> 0.05). The OR of CHD risk of CHD in the TGACTTCCTGGGTCAATG genotype was significantly higher than that of CHD risk of CHD-related death in the TGACTTCCGTCAATG genotype (< 0.05).
